研究了有机相中脂肪酶催化的炔丙醇酮乙酸酯的立体选择性醇解反应,考察了碱的种类、酰基受体和溶剂等对反应的影响.结果表明,以四氢呋喃为溶剂,CH_3OH为酰基受体,Lipase PLG脂肪酶为催化剂,Na_2CO_3为碱性添加剂,高底物浓度下40℃反应96h后,底物转化率和产物ee值分别达到49.5%和99.5%.碱的添加极大地提高了反应速度.
Kinetic resolution of (RS)-4-hydroxy-3-methyl-2-(2'-propynyl)-2-cyclopentenone (HMPC) acetate was carried out successfully by lipase-catalyzed alcoholysis in organic media. Using tetrahydrofuran as the solvent, methanol as the acyl acceptor, and lipase PLG as the catalyst, the conversion of substrate and the ee value of product were 49.5% and 99.5%, respectively, after 96 h reaction at 40 ℃ and high substrate concentration. It is noteworthy that basic additives can significantly promote the reaction.
