以α-单硬脂酸甘油酯的水解反应为指示反应,初步研究了琥珀酸二辛酯磺酸钠(AOT)-正庚烷-明胶微乳液凝胶固定化脂肪酶在有机相中的水解催化活性. 结果表明,明胶和AOT含量、R值(水与AOT的摩尔数之比)对酶催化活性均有影响,确定了较佳的凝胶组成:微乳液配比为m(正庚烷)∶ m(缓冲液)∶ m(AOT)=75∶ 9∶ 16,明胶质量分数为17%,R值为72;当酶含量固定,底物浓度大于0.10 mol/L时,脂肪酶可发挥最大催化活性;反应时间超过15 h,水解反应达到平衡.
