采用柠檬酸三钠还原法制备了胶体银用于标记羊抗人前白蛋白.在pH =7.0的Na2 HPO4- NaH2PO4缓冲溶液中,银标记前白蛋白抗体与前白蛋白抗原产生特异性结合,在聚已二醇和氯化钾作用下,导致体系在470 nm处的共振散射强度降低.当前白蛋白浓度在0.0667~2.668μg/mL范围内,随着前白蛋白原浓度的增加,在470 nm处的共振散射强度线性增强,方法的检出限为0.0421 μg/mL.,用于定量分析人血清中的前白蛋白,结果满意.
