保护剂溶液冻结及冻干特性的研究对生物材料保护剂的选择以及冷冻干燥工艺的设计具有重要的指导意义。选择聚乙烯醇(PVA)、海藻糖和叔丁醇作为锥虫的保护剂,采用差示扫描量热法研究了PVA/海藻糖/叔丁醇保护剂溶液的冻结特性,并对PVA/海藻糖/叔丁醇保护剂溶液进行了冷冻干燥实验,考察了不同冷冻条件对PVA/海藻糖/叔丁醇保护剂溶液冻结特性和冻干特性的影响。
The study of freezing and freeze-drying of protectant solutions is of great importance for the selection of protectants and the design of freeze-drying processes for the freeze-drying of biomaterials. In the present study, polyvinyl alcohol (PVA), trehalose and tert-butanol were selected as protectants for the freeze-drying of trypanosome, and they were used to prepare the aqueous protectant solutions. The freezing behavior of the aqueous protectant solutions of PVA, trehalose and tert-butanol was investigated by the differential scanning calorimetry (DSC), while the freeze-drying behavior was investigated by the freeze-drying system coupled by microscopy. The effects of freezing condition, especially freezing rate and tert-butanol concentration, on the behavior of freezing and freezing-drying of the aqueous protectant solutions were mainly investigated.
