通过插层技术合成了醋酸洗必泰-盐酸特比萘芬/蒙脱土(CA-TBNF/MMT)药物控、缓释纳米中间体,利用XRD对其结构进行了表征,结果表明CA-TBNF已进入MMT层间,MMT层间距从1.53nm增大到4.01nm.CA-TBNF/MMT在生理盐水中的体外缓释实验表明,吸附在MMT层板表面与插层到MMT层间的CA-TBNF,分别以一定的作用力方式与MMT作用,前24h的CA、TBNF快速释放,保证药效的及时性;而24h后释放缓慢,满足药物抗菌的持久性.由于CA、TBNF与MMT的静电作用,CA、TB-NF之间的协同作用以及MMT片层的阻隔效应,最终实现了药物的可控缓释.
The present paper focused on the intercalation of chlorhexidine acetate (CA) and terbinafine hydro-chloride (TBNF) into montmorillonite (MMT) as a sustained release drug carrier. The intercalation compounds were characterized by X-ray diffraction (XRD). The basal spacing of MMT increased from 1.53 to 2.97nm. In vitro release tests in 0. 9wt% NaCI solution at 37℃ showed a biphasic and prolonged release profile of CA and TBNF. The release rate depended on the electrostatic interactions between the drugs and MMT. The initial fast release quickly allowed establishment of a therapeutic dose,and the subsequent sustained release allowed mainte-nance of this does over a long period of time.
