对5种还原法溶解体系提取羊毛角蛋白的溶解率及大分子的空间结构等做了比较测试和分析.其中,TCEP/亚硫酸氢钠溶解体系溶解时间合理,且溶解率和质量分数高.FTIR测试结果表明,所提取角蛋白的α-螺旋结构和β-折叠结构的含量较原毛都有所下降,而无规卷曲的含量增加明显,但变化程度不一.DSC测试结果表明,提取的角蛋白的α-结晶的熔融峰温各不相同,说明溶解改变了角蛋白大分子的结晶情况,且工艺不同,结晶情况改变程度也不同.
