以紫外光度法监测胶原生物矿化的动力学过程,用液氮冻结法采集胶原生物矿化反应中不同时间状态点的样品,通过扫描电镜和能谱分析,研究胶原生物矿化过程中人工骨表面结构的变化及磷酸钙组成的递变过程.结果表明:胶原自组装过程先于磷酸钙矿化.胶原首先自组装成负载钙离子的板式结构,随后磷酸根在模板上进行磷酸钙矿化过程.在矿化过程中,钙、磷摩尔比由2:1过渡到1.89:1,逐渐趋向于羟基磷灰石组成,胶原矿化物依次经历了疏松的环状、层状和均匀稠密的网状等结构变化,证明了磷酸钙由无定形向晶形转化的推测,为胶原生物矿化过程中的模板作用提供了佐证.分析了胶原生物矿化的机制,解释了紫外吸收动力学曲线上的负峰现象.

The kinetic process of collagen biomineralization was monitored by ultraviolet photometry. The samples at different reaction states were obtained by liquid nitrogen freezing in the process of collagen biomineralization. The varieties of structure and composition of collagen biomineralization in reaction process were studied by scanning electron microscope (SEM) and energy spectrum (ES). The results indicated that the self-assembly of collagen occurred before calcium phosphate mineralization. Collagen firstly formed sandwich contained calcium ion through self-assembly, and then calcium phosphate formed on the sandwich templates. The mineral with the composition of hydroxyapatite was generated when the ratio of calcium to phosphorous changed from 2:1 to 1.89:1. The collagen biomineral structure had the evolution from orbicular to layer-shape, and finally to net with well-proportioned and dense structure. The assumption that the structure of calcium phosphate changed from amorphous to crystalline, and the template action of collagen in the process of biomineralization were proved. The mechanism of collagen biomineralization was analyzed, and the phenomenon of negative peak appeared in kinetic ultraviolet absorption curve of biomineralization was explained.
